Computational Screening Identifies GyrB Inhibitor Leads From Traditional Chinese Medicine Compounds

Published in Artificial Intelligence, Drugs development & discovery, Journals.

A Discover Chemistry study published on September 17, 2026 uses a computational pipeline to search a Traditional Chinese Medicine compound library for potential inhibitors of Escherichia coli DNA gyrase B. The research received new media coverage on October 4.

Peter Adeolu Adedibu and colleagues combined machine learning, pharmacophore filtering, docking and binding-energy calculations. They then explored structural modifications of lithospermic acid and used molecular dynamics to assess predicted interactions.

Lithospermic acid and a modified structure, Analog-1, emerged as computational leads. Favorable simulation scores against reference compounds do not demonstrate antibacterial potency or superiority to existing antibiotics.

The paper explicitly states that the findings need experimental validation. Predicted poor oral bioavailability is another important limitation. Synthesis, enzyme assays and bacterial testing will be necessary to determine whether the proposed molecules have practical value for antibiotic development.
