Iambic Submits IND for AI-Designed KIF18A Inhibitor IAM217

Published in Artificial Intelligence, Drugs development & discovery.

Iambic Therapeutics announced on October 5, 2026 that it submitted an Investigational New Drug application to the US FDA for IAM217. The proposed Phase 1/2 trial would evaluate the KIF18A inhibitor in solid tumours with chromosomal instability, including ovarian and triple-negative breast cancer.

According to Iambic, NeuralPLexer supplied structural insights during discovery without an experimental inhibitor-bound structure. Enchant and other property-prediction tools supported optimisation of potency, oral exposure, drug-interaction liabilities and brain penetration.

The company reports approximately 90% regression of established intracranial tumours in a preclinical brain-metastasis model. That result is nonclinical and does not establish effectiveness or safety in patients.

IND submission is a regulatory milestone, not FDA clearance or marketing approval. Trial initiation remains subject to FDA clearance. If cleared, IAM217 would become Iambic’s second programme to enter clinical testing, alongside its ongoing IAM1363 programme.
