Multi-organ chip review outlines what drug response models still need

Published in Drugs development & discovery, Journals, Tools.

A review published in Nature Biomedical Engineering on October 5, 2026, examines how human multi-organ-on-a-chip systems could help evaluate drug absorption, distribution, metabolism, excretion and toxicity.

These systems connect engineered tissues to model interactions between organs. The authors emphasize a central design problem: preserving each organ’s distinct biochemical and mechanical environment while allowing controlled exchange with other tissues.

The review discusses engineering approaches developed in single-organ models, their integration into multi-organ systems, scalable manufacturing and measurements using multiple readout methods. The aim is to produce more reproducible, informative data for drug evaluation.

The authors explicitly state that translation-ready multi-organ systems remain out of reach. Future connections to AI workflows are presented as a research direction, rather than a clinically validated AI platform.

This publication is a synthesis of existing research, not a new clinical trial or proof that organ chips can already replace all animal testing or accurately predict every patient’s response.
