Forward Therapeutics announced Phase 1 findings for FT2109, an investigational oral small molecule designed to inhibit soluble-TNF-driven TNFR1 signaling. The company says the findings support advancement into Phase 2 development.
The randomized, double-blind, placebo-controlled study involved healthy participants and evaluated single and multiple ascending doses. Forward reported a low adverse-event incidence comparable to placebo, with no serious adverse events or treatment discontinuations caused by treatment-emergent adverse events. Pharmacokinetic findings supported once-daily dosing.
Separate preclinical findings suggested anti-inflammatory activity while preserving membrane-TNF-mediated signaling and antibacterial responses in a challenge model. Those observations are not evidence of preserved infection protection in patients.
The company plans to present the clinical and preclinical data at ACR Convergence in November 2026. Healthy-volunteer safety findings do not establish efficacy in inflammatory disease or long-term safety, and the release does not announce that Phase 2 enrollment has begun.
