Lundbeck reported exploratory Phase Ib results for Lu AF28996, an investigational oral prodrug of a D1-like/D2-like dopamine receptor agonist, in advanced Parkinson’s disease. The company also announced that the first participant has been randomized in the Phase II DARE2 trial.
In the reported Phase Ib cohort, 34 participants received treatment for six weeks; 25 entered an optional extension. Mean daily ON-time without troublesome dyskinesia increased by 3.6 hours at week six and 3.4 hours at week 18. OFF-time decreased by 2.3 and 2.0 hours, respectively.
The study had no placebo or active comparator. These descriptive changes cannot establish treatment efficacy. Common adverse events included nausea, dizziness and falls; most were reported as mild.
DARE2 is a randomized, double-blind, placebo-controlled study targeting approximately 150 adults. Lu AF28996 remains investigational, with safety and efficacy not established.
