Seaport Therapeutics announced that the first patient has received GlyphAgo in an Australian phase 2a study involving adults with generalized anxiety disorder and sleep disturbance. The trial will investigate the candidate’s effects on sleep and anxiety symptoms.
GlyphAgo, also called SPT-320, is an oral prodrug of agomelatine. Seaport reported that an earlier phase 1 study in healthy volunteers found 6.8-fold higher bioavailability than unmodified agomelatine. That pharmacokinetic result concerns drug exposure; it does not demonstrate improved anxiety treatment.
The company expects topline results from the phase 2a study in early 2028. It also plans a global, randomized, double-blind, placebo-controlled phase 2/3 study in the first half of 2027, with results anticipated by the end of 2028.
This announcement marks the start of patient testing at this stage, not a positive efficacy readout. GlyphAgo remains investigational, and planned trial dates may change.
