GS030 Optogenetic Therapy: PIONEER Safety Results and Exploratory Visual Signals

Published in Drugs development & discovery, Journals, Tools.

GenSight Biologics and Paris’s 15-20 National Hospital announced publication of the phase 1/2 PIONEER results in the New England Journal of Medicine. The study evaluated GS030, an experimental optogenetic approach for advanced retinitis pigmentosa.

GS030 combines an injection of an AAV gene therapy into one eye with light-stimulating goggles. The gene therapy makes surviving retinal ganglion cells light-sensitive, aiming to bypass lost photoreceptors.

According to the announcement, six of ten participants showed improved light sensitivity in exploratory testing. Four of eight participants completing behavioral testing improved on certain visual tasks while wearing the goggles. Most ocular adverse events were mild or moderate; one severe event resolved within minutes after treatment. No systemic adverse event was attributed to the study drug or injection.

PIONEER was an open-label dose-escalation study focused on safety. Its small size and exploratory assessments do not establish efficacy. Larger confirmatory studies are needed, and GS030 has no marketing authorization.
