Encaleret: CALIBRATE update reports symptom and bone-turnover changes in ADH1
Published in Diseases!, Drugs development & discovery.
Published October 11, 2026. BridgeBio reported additional Phase 3 CALIBRATE findings for investigational encaleret in autosomal dominant hypocalcemia type 1 (ADH1) at the ASBMR meeting on October 11.
According to the sponsor’s 24-week presentation, endogenous parathyroid hormone reached at least the lower limit of normal in 91% of encaleret recipients versus 0% on standard care. Markers of bone formation and resorption also increased.
In the participant-reported symptom analysis, improvement in at least one symptom was reported by 100% of encaleret recipients versus 46% receiving standard care. Reported improvements included fatigue, muscle cramps and tingling. These percentages describe symptom reports, not disease cure.
Limitations: This is a sponsor-reported conference update. Symptom reporting and changes in bone-turnover markers do not alone establish long-term skeletal benefits. Encaleret remains investigational; regulatory review is not approval.
