Tovinontrine Phase 2 trials report mixed heart-failure biomarker results
Published in Diseases!, Drugs development & discovery.
Published October 11, 2026. Cardurion announced Phase 2 results for its investigational oral PDE9 inhibitor tovinontrine (CRD-750) at the Heart Failure Society of America meeting on October 11.
In Cycle-1REF, the company reported that 529 participants with heart failure with reduced ejection fraction were randomized to three doses or placebo alongside standard therapy. NT-proBNP, a heart-failure biomarker, decreased over 12 weeks at two of the three doses, with the largest reduction at 2.5 mg twice daily.
The separate Cycle-2PEF trial randomized 277 participants with preserved ejection fraction to 50 mg twice daily or placebo. It found no significant NT-proBNP change at that dose. Cardurion described tolerability and safety as favorable in both studies and plans further development.
Limitations: These are sponsor-reported conference results. The announcement does not provide the numerical effect sizes or detailed adverse-event rates needed for independent assessment. A biomarker reduction alone does not establish fewer hospitalizations or longer survival.
