AI-designed menin inhibitor CHM-029 enters human testing for acute myeloid leukemia

Published in Artificial Intelligence, Diseases!, Drugs development & discovery.

Published October 12, 2026. CHARM Therapeutics announced on October 7 that the first patient had received CHM-029 and the first cohort was fully enrolled in its phase I/II trial for acute myeloid leukemia.

CHM-029 is an experimental menin inhibitor designed using DragonFold, the company’s AI platform for modeling protein–ligand interactions. The aim is to retain activity against mutations that can cause resistance to earlier menin inhibitors.

The trial, NCT07751991, will evaluate safety and efficacy in relapsed or refractory leukemia with an NPM1 mutation, KMT2A rearrangement or NUP98 rearrangement. Initial data are expected in 2027.

CHARM reports activity against resistance mutations and tumor regression in preclinical models. Those laboratory findings support clinical testing but do not establish patient benefit.

Scope: First dosing marks the start of human evaluation. No clinical response or comparative safety results were reported in this announcement, and CHM-029 remains investigational.
