Caspian and Kura outline KO-7246 path toward a diabetes proof-of-concept trial

Published in Diseases!, Drugs development & discovery.

Publication date: October 13, 2026.

Caspian Therapeutics and Kura Oncology outlined a development strategy for KO-7246, an oral menin inhibitor intended to increase the body’s insulin-producing capacity. The candidate remains in IND-enabling development, with a planned phase I proof-of-concept study in people with diabetes.

The October 13 update summarizes findings previously presented at EASD. The companies reported beta-cell expansion and improved glucose control in diabetic mice, sustained responses after treatment withdrawal in responding insulin-deficient rats, and beta-cell proliferation in human islet microtissues. These are preclinical observations, including experiments on human tissue outside the body.

The initial clinical objective is to characterize safety, tolerability and drug exposure, while looking for changes in endogenous insulin production. Stimulated C-peptide measurements during treatment and follow-up are proposed to help assess biological response.

Limits: the release does not report treated-patient outcomes or clearance to start the planned trial. Animal responses and human tissue proliferation do not establish safe, durable regeneration in people or a diabetes cure. Responses in rats appeared dependent on remaining beta-cell capacity.
