Recurring gliomas follow distinct genetic paths, longitudinal study finds

Published in Bioinformatics, Brain/Neurology, Diseases!, Journals.

Published October 15, 2026.

A Nature study published October 14 followed genetic changes in tumor samples from 183 patients with IDH-mutant glioma. The GLASS Consortium analysis compared samples collected at different surgeries to investigate how astrocytomas and oligodendrogliomas evolve as disease progresses.

Recurrent oligodendrogliomas acquired new driver mutations, including changes involving PI3K and NOTCH1. Astrocytomas more prominently accumulated changes in DNA copy number and chromosomal instability. Several acquired alterations were associated with worse outcomes.

The findings suggest that these two tumor types may require different strategies for studying treatment resistance and designing future therapies.

Evidence limits: This genomic analysis did not test a new treatment. Its associations do not establish that changing care according to these findings improves survival. The cohort also required patients to survive to another surgery, creating a selection bias that limits how broadly the results apply.
