Estradiol–Autophagy Pathway Linked to Vascular Remodeling in Hypertensive Rats

New preclinical research links estradiol signaling to a cellular recycling pathway that may help limit blood-vessel changes associated with hypertension. Miao-Jin Ji and colleagues published the study in Communications Biology on October 17.

In spontaneously hypertensive rats, removal of the ovaries worsened hypertension, while estradiol supplementation reduced blood pressure and vascular remodeling. The researchers investigated GRK2, a protein involved in signaling that promotes vascular smooth muscle cell proliferation and migration.

Their experiments indicate that estradiol lowers GRK2 levels, with corresponding reductions in β-arrestin2 recruitment and ERK signaling. Blocking either autophagy—the cell’s recycling machinery—or estrogen receptor alpha prevented estradiol’s effect on GRK2 degradation.

The findings identify a mechanism worth investigating as a possible therapeutic target. They do not establish that menopausal hormone therapy prevents cardiovascular disease or should be prescribed for hypertension. Animal and cellular results require clinical validation, including assessment of treatment risks and benefits in people.
